David Scott Mustaine , nasceu em 13 de setembro de 1961 na cidade de La Mesa, Califórnia, Estados Unidos, filho mais novo de Emily e John Mustaine.
Os pais de Dave eram testemunha de Jeová. Seu pai, John, era alcoólatra e Dave era frequentemente espancado por ele. Após seus pais se divorciarem, Dave, sua mãe e suas irmãs passaram e se mudar frequentemente para evitar contato com o pai violento. Aos quinze anos, ele alugou um apartamento para si e passou a traficar drogas para sobreviver.
Uma de suas clientes estava quase sempre sem dinheiro, mas trabalhava numa loja de discos, e oferecia a Dave álbuns de artistas como Iron Maiden,Black Sabbath e Judas Priest para saldar as dívidas, o que o ajudou a moldar sua preferência pelo heavy metal (suas bandas favoritas são AC/DC e Kiss). No final da década de 1970, Dave começou a aprender guitarra — notavelmente com uma B.C. Rich — e juntou-se a uma banda chamada Panic por um curto período de tempo
No ano de 1981, Mustaine conheceu Lars Ulrich através de um anúncio num jornal chamado Recycler, e entrou para o Metallica, juntamente com Ron McGovney no baixo.
Mustaine diz em sua autobiografia que sua primeira expulsão do Metallica aconteceu no verão de 1982, num ensaio na casa de Ron. Quando chegou, sua cachorra saiu de seu carro e pulou no capô do carro de Ron, um Pontiac GTO. James Hetfield acertou um chute forte na cachorra, que Dave não deixou passar.
Quando entraram na casa de Ron começaram as ameaças e insultos, até Dave dizer a James para ficar quieto se não daria um soco em sua boca. Ron tentou defender James, e Dave acabou acertando um soco em James e uma cotovelada em Ron, que caiu sobre os equipamentos. Um amigo de Dave, Rick Solis, interveio e separou a briga. Naquela hora James expulsou Dave. Porém Dave voltou a ensaiar com a banda menos de vinte e quatro horas depois, após pedir desculpas a todos.
Sua saída definitiva do Metallica aconteceu em 11 de Abril de 1983. Depois de fazerem dois shows, Dave acordou de manhã com a equipe toda em sua volta com suas malas prontas. Dave voltou para a Califórnia de ônibus sem um único dólar, em uma viagem de quatro dias.
Mesmo saindo do Metallica, Dave criou grande parte das letras, riffs e solos do primeiro álbum da banda, Kill 'Em All e contribuiu em duas músicas do segundo Ride the Lightning.
Após ser demitido do Metallica, Dave criou a banda Megadeth, com o baixista David Ellefson, o guitarrista Chris Poland e o baterista Gar Samuelson. Com esta formação lançaram já em 1985 o álbum Killing Is My Business... And Business Is Good!, bastante aclamado pelo público e mídia.
Após lançarem Peace Sells... But Who's Buying? em 1986, Chris Poland e Gar Samuelson foram despedidos da banda por Mustaine, sendo substituídos pelo guitarrista Jeff Young e pelo baterista Chuck Behler. Essa formação lançou um novo álbum chamado So Far, So Good... So What!, com destaque para a música "In My Darkest Hour".
O relacionamento de Mustaine com o restante do grupo nunca foi muito bom. Ele sofria de ataques de overdose, e foi motivo de polícia muitas vezes por isso. Um dia a polícia o prendeu por um motivo grave, oito tipos de drogas injetáveis distintas. Após esse dia ele teve que ser internado em uma clínica de reabilitação.
Meses depois Mustaine se dizia um novo homem e reformulou a sua banda, com Marty Friedman na guitarra, Nick Menza na bateria e David Ellefson no baixo.
Esta formação lançou o álbum Rust in Peace, que foi sucesso de vendas em todo o mundo, e considerado por Mustaine como o melhor trabalho do grupo e considerado por muitos um dos maiores discos da história do metal. Em 1992, lançaram Countdown To Extinction, com destaque para a faixa "Symphony Of Destruction".
O próximo álbum, Youthanasia, foi lançado em 1994; a faixa "A Tout Le Monde" foi, por cinco meses, uma das mais tocadas nas rádios dos Estados Unidos. Em 1995, eles lançam o álbum Hidden Treasures.
Em 1997 o Megadeth lança Cryptic Writings, um álbum que pretendia retornar a banda às suas origens, com músicas que lembram toda a carreira da banda: "She-Wolf", "Trust", "Secret Place", "Vortex", "FFF", "Almost Honest", e "The Disintegrators".
Nesse mesmo ano Nick Menza foi despedido da banda por Mustaine, por causa de um tumor no joelho, sendo assim, ele foi substituído por Jimmy DeGrasso.

Em 2004, Mustaine já recuperado volta a ativa com o Megadeth, e no mesmo ano de retorno, a banda lança um novo álbum chamado The System Has Failed para muitos um dos melhores da história do grupo, com os integrantes Chris Poland (guitarra solo), Vinnie Colaiuta (bateria) e Jimmie Lee Sloas (baixo). Era para esse ser um álbum solo de Mustaine, mas o mesmo mudou de ideia, e depois da gravação resolveu fazer uma reformulação da banda.
Poucos dias depois, Mustaine anuncia uma nova formação, composta pelos irmãos Glen e Shawn Drover, e o baixista James MacDonough (ex-Iced Earth). Fazem uma turnê pelo mundo inteiro, em 2004 e 2005, inclusive no Brasil.
Desde o retorno de 2004, o Megadeth lançou 5 álbuns de estúdio: The System Has Failed (2004), United Abominations (2007), Endgame (2009), Thirteen (2011) e Super Collider (2013) além da coletânea Greatest Hits: Back to the Start (2005) e o DVD duplo Arsenal of Megadeth (2006).
Dave e o Megadeth fizeram a abertura do show de retorno da banda Black Sabbath em 2013 , e eles continuam a tocar firmes e fortes até hoje . E para a surpresa de muitos o nosso guitarrista brasileiro ( Kiko Loureiro foi contratado para se juntar ao Megadeath , pois o próprio Dave diz ter se maravilhado com os acordes dele .

David Scott Mustaine was born on September 13, 1961 in La Mesa, California, United States, the youngest son of John and Emily Mustaine.
Dave's parents were Jehovah's Witness. His father, John, was an alcoholic and Dave was often beaten by him.
